purpose demo BOTH myelin and nissl substance
luxol fast blue principle stains due to lipoproteins. it is an adic-base reaction that forms salt. the base of lipoprotein replaces the base of the dye.
cresyl echt violet principle Nissl susbstance is basophilic and stains sharply with basic aniline dyes. Varying the PH and differentiation, nissl substance and nuclei will stain or only nissl substance will demo.
fix formalin

qc spinal cord or medulla
reagents in procedure luxol (stain myelin), lithium carbonate (diff'r), 70% alcohol (diff'r), cresyl echt violet (counterstain), alcohol (diff'r)
results myelin - blue, nissl/nuclei - violet/rose (purplish)
issues with this stain are typicall the result of which part the cresyl echt violet causes typical issues
if acetic acid isn't added to cresyl echt what will occur the background will get a diffuse violet stain (entire slide will be purplish)
if the cresyl echt violet isn't heated what will occur there will be decreased staining of the nissl substances
what does the cresy echt violet do for this stain intensifies the staining of the myelin sheath
